500mg NMN on empty stomach. Restores NAD+ - the fuel that AMPK-activated pathways run on. Daily throughout and between MOTS-c/AICAR cycles.

MOTS-cTier B

10 mg

10mg subcutaneous injection 3-5x per week during active cycle (4-8 weeks on). Morning timing aligns with natural metabolic signalling rhythms. Do not use on same day as AICAR - alternate days.

50mg subcutaneous injection on MOTS-c rest days (2-3x per week). NOT the same day as MOTS-c. Monitor blood glucose before and 60 min after injection. Start at 25mg first week to assess tolerance.

Protocol Overview

The Metabolic Optimizer operates at three levels of cellular energy regulation simultaneously:

NMN (daily) -- NAD+ Restoration NAD+ is the electron carrier that both AMPK pathways and sirtuin pathways depend on. Without adequate NAD+, the MOTS-c and AICAR components cannot exert their full downstream effects. NMN is the foundation -- run it daily throughout the protocol.

MOTS-c (3-5x per week) -- Mitochondrial Signalling MOTS-c is produced by mitochondria themselves in response to metabolic stress. Exogenous MOTS-c activates AMPK through the mitochondrial pathway, improves insulin sensitivity, and mimics the metabolic adaptations of endurance exercise. It additionally regulates nuclear gene expression through direct nuclear translocation -- a unique mechanism among AMPK activators.

AICAR (2-3x per week, alternate days) -- Direct AMPK Activation AICAR is the most potent direct AMPK activator available outside of pharmaceutical intervention. Where MOTS-c activates AMPK via mitochondrial signalling, AICAR activates it directly through AMP mimicry. The combination of two independent AMPK-activating pathways (plus the NAD+ provided by NMN) produces comprehensive metabolic reprogramming.

Why Alternate Days for MOTS-c and AICAR?

Both MOTS-c and AICAR activate AMPK, but through different mechanisms. Running both on the same day significantly increases the risk of hypoglycaemia and may over-activate stress response pathways. Alternating them maximises coverage while maintaining a manageable safety profile.

Example weekly schedule:

  • Monday: NMN + MOTS-c
  • Tuesday: NMN + AICAR (+ glucose monitoring)
  • Wednesday: NMN + MOTS-c
  • Thursday: NMN (rest from injectables)
  • Friday: NMN + MOTS-c
  • Saturday: NMN + AICAR (+ glucose monitoring)
  • Sunday: NMN only (recovery day)

Required Monitoring

Before starting: Fasting glucose, HbA1c, complete blood count, liver enzymes

During AICAR days: Blood glucose at baseline and 60 minutes post-injection (hypoglycaemia risk is real)

At cycle end (8 weeks): Repeat full panel + HbA1c

WADA Notice

AICAR is prohibited by WADA in competitive sport, both in and out of competition. Athletes subject to anti-doping testing must not use this compound.
